Chemistry of Heterocycles

Assessing Algerian Ilex aquifolium L.'s phenolic compounds, antioxidant and enzyme inhibitory properties to find a new natural remedy for Alzheimer's, Diabetes and Skin Diseases

This research explored the impact of methanolic extracts from the
fruits and leaves of Ilex aquifolium L., a rare Algerian species, on
several enzymes, focusing on tyrosinase, α-amylase, α-glucosidase,
and cholinesterases, as well as their antioxidant potential in vitro
assays, including phosphomolybdenum, DPPH, ABTS, CUPRAC,
FRAP, and MCA. HPLC-DAD analysis identified chlorogenic acid, rosmarinic
acid, hesperidin, and quercetin as predominant phenolic
components. Both extracts exhibited notable antioxidant properties,
up to 250.08 mg TE/g for fruits and 237.08 mg TE/g for leaf
extracts. Enzyme inhibitory assays demonstrated significant inhibition
against AChE and BChE with maximum activities of 2.34 mg
GALAE/g in leaves and 4.32 mg GALAE/g in fruits. Additionally, considerable
tyrosinase inhibition was observed in leaves at 39.69 mg
KAE/g. These findings highlight I. aquifolium L. as a valuable source
for investigating natural antioxidants and enzyme inhibitors, particularly
for possible cosmetics and food additives applications.

Biological effects and chemical characterization of Ilex aquifolium L. extracts: A new source of bioactive constituents

This study gathers information about the effects of methanolic extracts from fruits and aerial parts of Ilex aquifolium L. on selected enzymes (cholinesterases, alpha-amylase and alpha-glucosidase and tyrosinase ) as well as their antioxidant capacities in relation with their phenolic composition. The chemical composition was assessed by determining total phenolic and flavonoid content as well as individual phenolic compounds by HPLC-DAD. Moreover, antioxidant abilities of the investigated extracts were tested by using different assays including free radical scavenging (DPPH and ABTS), reducing power (CUPRAC and FRAP), phosphomolybdenum and metal chelating, overall fruits being indicated as a source of antioxidant compounds. Furthermore, interactions between dominant compounds from extracts and selected enzymes were investigated by molecular modeling studies in order to explain at a molecular level the interactions between selected compounds and active sites of the enzyme.

Phenolic contents and antioxydant activity of the methnanolic extract of Ilex aquifolium species

Ilex aquifolium is a medicine plant used in traditional medicine for diuretic, purgative and tonic properties as well as for the treatment of liver, stomach and intestinal cancers. This study aimed to investigate the phenolic constituents and to evaluate antioxidant activity of Ilex aquifolium L.The Determination of Total Phenolic compounds was investigated by Folin- Ciocalteu method. The results revealed the presence of 126.7 mg Gallic acid / g leaves extract. Moreover, flavonoids content were dertminated using aluminum trichloride method, which reveals a concentration of 60.14 mg Quercetin / g leaves extract The antioxidant activity was determined using DPPH method (Free Radical Scavenging technique). The results showed a significant antioxidant activity with an IC50 equal to 0.87mg .

Antioxidant and antibacterial activities of the ethanolic extract of Artemisia herba alba species

The objective of this work was to evaluate the antioxidant and antibacterial activities of the ethanolic extract of Artemisia herba alba species collected from Khenchela region (Hammam knif). Total phenols, Flavone and flavonol were quantified spectrophotometrically. The antioxidant activity of this plant was estimated by the DPPH free radical scavenging and total antioxidant capacity. The results showed that the studied species had a very interesting activity. Antibacterial tests were determined using ATCC reference bacterial strains. The ethanolic extract was found to be ineffective for the majority of the tested bacteria.

Antioxidant capacity and total phenolic content of the leaves and the bark of Fraxinus angustifolia spiecies

The objective of this work is to evaluate the phenolic content and antioxidant activity of methanolic extracts of leaves and barks of an autochthonous species namely Fraxinus angustifolia Vahl. ssp.(Oleaceae). Total phenols and flavonoids were quantified spectrophotometrically. The antioxidant activity of this plant was estimated by the DPPH free radical scavenging and total antioxidant capacity. The total phenolic and flavonoid contents were 160.1±0.8 mg GAE/g of extract and 15.2±2.1 mg QE/g of extract, respectively. The methanol extract showed inhibition, above 70% for the DPPH scavenging ability and lower than 50 % for ABTS assay, for 20 µg/ml of extract.

A New coumarin-secoiridoid from the stem bark of the Algerian medicinal plant Fraxinus xanthoxyloides

This paper deals with the isolation and structure identification of the new compound isofraxisecoside, a coumarin- secoiridoid diglucosides along with nine known compounds. To the best of our knowledge, isofraxisecoside, is the third example of a natural compound consisting of one coumarin glucoside unit linked to a secoiridoid moiety of oleoside type and no phytochemical investigation has been carried out on the Algerian species Fraxinus xanthoxyloides (G.Don) Wall. ex A.DC. The molecular structures of the isolated compounds were mainly elucidated by the use of NMR techniques 1D and 2D (1H, 13C, DEPT, HSQC, HMBC and ROESY), high resolution mass spectrometry (HR-ESI-MS), measurement of the optical rotation [α]D20 and by comparison with the literature data.

Chemical Constituents And Antioxidant Activity Of Fraxinus Xanthoxyloides Leaves Extract

Introduction :
F. xanthoxyloides (G.Don) Wall.(Oleacea family) have been used in folk medicine in different parts of the world for their diuretic, mild purgative effects and for the treatment of constipation.
Objectives :
The aim of the present work is the investigate the constituents of the leaves and to estimate the antioxidant potential.
Materials and Methods :
The methanolic extract of the leaves of F. xanthoxyloides was submitted to subsequent purification steps by using different chromatographic techniques, mainly the law pressure liquid chromatography (LPLC). The molecular structures were established mainly by 1H-NMR, 13C-NMR. The total phenolics and flavonoids were quantified spectrophotometrically. DPPH and ABTS assays were employed to evaluate the antioxidant activity.
Results :
Ten compounds have been isolated from the leaves including three secoiridoid glucosides of oleoside type, three coumarins, two flavonoid diglucosides and two phenylethanoid glucosides. The total phenolic and flavonoid contents were 22.1±0.8 mg GAE/g of extract and 22.2±2.1 mg QE/g of extract, respectively. The methanol extract showed inhibition, above 50% for the DPPH scavenging ability and lower than 50 % for ABTS assay, for 200 µg/ml of extract.
Conclusion :
This study constitutes a preliminary work to evaluate the content in polyphenols and the antioxidant activity of the extract F. xanthoxyloides leaves. These promising results pave the way for further detailed studies of their chamical composition and finding of the active components.
