Fatima Gareche, Oum Elkheir Belhaddad and Abd el-Hamid Guelil. Synthesis, characterization and biological studies of metal (II) Complexes of n-[(pyridin-2-ylimino) methyl]-tetra-o-acetyl-β-d-glucopyranose

In the therapeutic field, several researchers are directed towards the discovery and the synthesis of new molecules likely to respond to patient expectations. In this work, we have synthesized the ligand N-[(pyridin-2-ylimino)methyl]-tetra-O-acetyl-β-D-glucopyranose (L). The latter was complexed metals of transition (II) such as Manganese, Nickel and Zinc. The condensation of 2-pyridinecarboxaldehyde with 1,3,4,6-tetra-O-acetyl-2-amino-2-deoxy-β-D-glucopyranose hydrochloride in absolute ethanol in a molar ratio of 1 : 1 led to the production of (2-imine). This imine was used as a monodentate ligand to coordinate metal salts of Mn(II), Ni(II) and Zn(II), the ratio of (Metal – Ligand) was 2 : 1. All the compounds obtained were characterized by UV-visible spectroscopy. The electroless nature of the metal complexes was confirmed by molar conductance studies. The synthesized (2-imine) and their metal complexes were tested for their antibacterial activities against Escherichia coli and Staphylococcus aureus. In another hand, antioxidant activity was tested by assay with the 2,2-Diphenyl radical-1-picrylhydrazyl at different concentrations. All the complexes showed significant antibacterial activity and higher antioxidant activity than the corresponding imine. The considerable results found proved that imine and their complexes are bioactive in nature.

Phytochemical characterization, biological screening and corrosion inhibition of mild steel from extracts of Juniperus oxycedrus L

The aim of this research was to study the effect of solvent polarity and extraction technique of phenolic compounds from Juniperus oxycedrus L. on antioxidant and antibacterial activity. Also, this paper aims to evaluate the anti-corrosive effect of aqueous extracts. In order to evaluate the effect of solvents on the bioactive
compound’s extraction efficiency, the samples were prepared with different extraction procedures. The methanolic and ethanolic extracts were prepared by using Soxhlet and maceration extraction and aqueous extracts were obtained by decoction and infusion methods. Moreover, six fractions (petroleum ether, chloroform, ethyl acetate, 1-butanol and aqueous) were prepared by partition of ethanolic extract. All the extracts were subjected to preliminary phytochemical screening; the results show the presence of flavonoids, tannins, alkaloids, triterpenes and steroids. The study of the effect of extraction methods revealed that the decoction is the most suitable for the extraction of phenolic compounds. Moreover, depending on the solvent studied, it was noted that the ethanol showed a high level of flavonoids and the high antioxidant capacity. The extracts were also tested for their antibacterial activity in vitro against two bacterial strains (Escherichia coli and Staphylococcus aureus) using the disc diffusion method. The results of antibacterial showed that the highest activity was attributed to ethanolic extract with a maximum zone of inhibition of 13 mm against Staphylococcus aureus. The corrosion inhibition effect of the aqueous extracts in mild steel in 1 M HCl solution showed that the extracts inhibit effectively corrosion of mild steel.

Etude comparative de l’utilisation de l’or bleu et/ou arthrospira platensis dans une formulation cosmétique

La Spiruline " Arthrospira platensis a été le sujet de nombreuses études dans plusieurs pays. Le cout élevé des éléments minéraux utilisés dans les cultures en masse de cette algue reste un problème sérieux et la nécessite d’avoir recours à une source minérale s’impose : les eaux de la région de M’sila (Algérie) peuvent être une solution adéquate vu leur richesse en éléments minéraux. En plus, la Spiruline " Arthrospira platensis est intérêt thérapeutique grâce à ses pigments naturels appelés phycobiliprotéines, et leur potentiel bénéfique a été expérimentalement prouvé in vitro et in vivo pour traiter certaines pathologies. La phycocyanine (PC) est un complexe pigment-protéine constitue environ 50 % de phycobiliprotéine et représente jusqu'à 20 % du poids sec de la Spiruline. Elle joue un rôle bénéfique dans la santé humaine grâce à ses propriétés pharmacologiques, biologiques et thérapeutique potentielles. En effet, plusieurs études épidémiologiques ont mis en évidence l’effet bénéfique de la PC sur la santé humaine d’une part ses différentes propriétés: antioxydantes et anti-inflammatoires, immunomodulantes, immunostimulantes, anti-cancérigènes, antidiabétiques, athéroprotectives, et en cosmétiques.En raison des multiples utilisations de la spiruline et PC, nous avons été encouragés à faire certains cosmétiques qui jouent un rôle important dans le traitement des rides et des taches avec effet efficace sur les personnes volontaires qui ont testé les savons et les crèmes préparés .

Stabilisation de la Curcumine approches structurale : Preuve de Concept et investigation de la composition idéale des formulations.

La Curcumine, principe actif majeur du curcuma, est un composé polyphénolique présent dans les racines et le rhizome de l’épice (Curcuma longa L.). Dans la médecine traditionnelle, elle peut être administrée sous forme de plante entière, d’extraits concentrés, de curcuminoïdes ou de Curcumine seule. La Curcumine est plébiscitée dans toutes les thérapies holistiques pour ses bienfaits sur la sphère articulaire, cardiaque, hépatique, digestive, cutanée, immunitaire et nerveuse. Ses mécanismes d’action sont complexes et ubiquitaires, et sa capacité antioxydant la rend exceptionnellement efficace pour lutter contre les effets du vieillissement. Elle est très bien tolérée.
Cependant, l’utilisation de Curcumine est généralement limitée en raison de leur faible solubilité et stabilité et la biodisponibilité.
Les travaux réalisés dans cette invention ont apporté des résultats encourageants sur l’intérêt des CDs et PEG, FV dans les formulations alimentaires ou médicamenteuses de Curcumine. Le choix de cet Curcumine a été guidé par leur homologie structurale et leur propriété biologique et thérapeutique intéressante.

Synthesis, Characterization, and Biological Investigation of Transition Metal (II) Complexes Based on 2-Alkyl-2-Oxazolin-α-D-glucopyranose Modified Derivatives

A transition metal complexes based on Fe(II), Co(II), Ni(II), and Zn(II) were developed in this study, utilizing ligands derivatives from 2-alkyl-2-oxazolin-3,4,6-tri-O-acetyl-1,2-dideosy-α-D-glucopyranose derivatives. The elemental analyses suggested that the stoichiometry is (1:2) [Metal:(LDn)2]. The IR data confirmed the binding between the metal ion and the ligands. The crystallinity of the complexes formed was confirmed by the X-ray diffraction. The non-electrolyte nature of metal complexes was confirmed by molar conductance studies. The thermal study suggested the presence of coordinated water molecules in the complexes based on LD4 [M(LD4)2.(OAc)2.xH2O]. The synthesized complexes and their corresponding ligands were tested for their antimicrobial activities against bacteria (Escherichia coli and Pseudomonas aeruginosa [Gram negative]) and (Staphylococcus aureus and Streptococcus pneumonia [Gram positive]). The complexes [(Zn(LD4)2.(OAc)2.2H2O] and [Ni(LD4)2.(OAc)2.4H2O] showed significant antibacterial activity compared to the corresponding ligands. The dosage with the radical DPPH at different concentrations for the complexes [(Ni(LD1)2.(OAc)2.4H2O] and [Ni(LD2)2.(OAc)2.4H2O] showed superior antioxidant activity than the corresponding ligands. The considerable results found proved that the ligands and their complexes are bioactive.

Water compatibility of M’sila region (South-East Algeria) for cultivation of Spirulina & evaluation of methods of extraction of Phycocyanin and investigation of stability by β-Cyclodextrin

Spirulina (Arthrospira platensis) is a microalga that has existed for more than three billion years. It belongs to the family of cyanobacteria. It is extraordinary, rich in nutrients such as proteins, carbohydrates, lipids, vitamins, and minerals. In addition, Spirulina is rich in phycocyanin, a blue protein pigment. It can be used as a dye in food, pharmacology, and cosmetics. Our work focused first on the compatibility of the waters of M’sila region (southeast Algeria) for the cultivation of Arthrospira platensis. Then, we evaluated several methods of extracting phycocyanin to be able to determine the optimal method in terms of yield. We have succeeded in developing a new extraction method coupled with maceration in a glycerol-water mixture with a molar ratio (8/2) assisted by ultrasound with a yield of 52.93 mg/g. The excellent results obtained may be due to the salinity of the waters of the region, to the used nutrient culture medium and/or the climate change of the region. We have formed an inclusion complex between phycocyanin and β-Cyclodextrin to keep it and make it more stable. The encouraging results allow Algeria to gain a foothold on the world market as a producer of blue gold.

In this Work, we were interested on the synthesis of D-glucosamine derivatives. Within two steps; Acylation of the 1,3,4,6-tetracetyl-amino-2-deoxy-β-D-glucosamine hydrochloride (2) and the regioselective substitution by thiophenol. Given that this compound has very poor aqueous solubility and to ameliorate this, we have synthesized an inclusion complex between derivative (5) and β-CD, the preparation of the complex was carried out by two different methods (Physical mixture
method and Solvent evaporation method).The results of this work were confirmed by structural characterization by spectroscopic methods 19F NMR, 1H NMR and IR.Finally we studied the solubility of derivative (5) in an aqueous solution containing various concentrations of β-CD by UVVisible spectroscopy which was deducted enhancement of the derivative (5) solubility in the presence of β-CD.

Résumé sur les bases de la thermodynamique

Le terme « thermodynamique » vient de deux mots grecs : thermos (le feu) et dunamicos (la puissance). Cette discipline apparaît donc comme la science qui traite des relations entre les phénomènes thermiques et les phénomènes mécaniques. Si cette conception correspond bien aux travaux primordiaux réalisés aux XVIIIe et XIXe siècles (machines à vapeur de Newcomen, Watt, Papin et Carnot, moteurs thermiques de Otto, Diesel…etc.), de nos jours ce rôle initial est largement dépassé. Cette science s’applique dans tous les domaines en raison de la portée universelle de ses trois principes. En effet, un énoncé dans un domaine scientifique quelconque qui contredirait les principes de la thermodynamique serait immédiatement mis en défaut. La thermodynamique apparaît donc comme « la science des transformations de l’énergie, de la matière et des états d’équilibre ».

Synthèse, Structure, Réactivité de Dérivés Phosphorylés Hétérocycliques Contenant la D-Glucosamine. Etude du Complexe d’Inclusion et Évaluation Biologique

This thesis revolves around several axes in the field of the synthesis of the new original bioactive compounds. Firstly, we studied the regioselective deprotection reaction to obtain derivative (4) which is used as a precursor for the synthesis of potentially active compounds (carbamates and / or imines), we then studied the reactivity of the anomeric carbon C-1 of the latter by selective deprotection and then by hydrolysis to regenerate the alcohol at the α position. In the last step, we used Wang's condensation conditions and Ferry’s DMAP addition study to obtain the phosphorylated bicyclics with excellent diastereoselectivity. Yields are good in most cases. All the compounds synthesized were identified by the usual spectroscopic methods (NMR, IR, MS and UV-Visible), and for some of them additional analyzes were carried out (elemental analysis).
On the other hand and after evaluating Lipinski's rule which gives us positive information on the pharmacokinetic (ADME) and bioactive properties, we studied the solubility of the derivatives (2b, 5a, 5b, 5c, 7, 6, 7a, 9b and 6a) and the stability of the derivative (8a) in aqueous media at different pH (4.0, 6.9 and 8.5) by the formation of an inclusion complex based on β-CD (β-CD : derivatives) with a molar ratio 1/1. The results show that β-CD is able to encapsulate the studied derivatives, increase their solubility’s and improve their stabilities. The study of the biological activity suggests that the inclusion complexes formed can be considered as promising tools for the optimization of pharmaceutical formulations because the encapsulation has retained the antibacterial and antioxidant properties of the derivatives.

A novel approach toward the synthesis of 1,3,4,6-tetra-OAcetylazomethine based on β-D-glucosamine

A novel method was developed for synthesizing a series of 1,3,4,6-tetra-O-acetylazomethin-b-Dglucosamine
(5) starting from hydrochlorid β-D-glucosamine (1) with three different aldehydes in the
presence of 15% sodium hydroxide as catalyst. In these reactions 1/1, the condensation product as
half-unit ligand was obtained. The attractive features of this protocol are use of an inexpensive
catalyst, operational simplicity, short reaction times, easy handling, and good yields. Mild conditions,
simplicity and easier work-up are the main advantages of this method. In parallel, in this work we
succeed to separate the two diastereoisomers of products obtained (5a) which are characterized by
Visible UV and separated by crystallization to give (E)-isomer and (Z)-isomer.

Preparation, Characterization, and Biological Study of a New Inclusion Complex of 1, 3, 4, 6-Tetra-O-acetyl-2-deoxy-2-[benzyliden (amino)]-beta-D-glucosamine and beta-Cyclodextrin

In this study, an inclusion complex based on beta-Cyclodextrin (CD) and 1,3,4,6-tetra-O-acetyl-2-deoxy-2-[benzyliden(amino)]-eta-D-glucosamine (imine) were prepared in order to modify imine's solubility and improved its stability. The inclusion complex was obtained by equimolar quantity 1:1 under microwave irradiation and characterized by X-ray diffraction and melting point. The effect of pH on the formation of the inclusion complex in aqueous medium buffered at 25 degrees C was examined. Spectrophotometric measurements were carried out to visualize the interaction between imine and beta-CD in the complex in the liquid state. The results obtained showed that the complexation was favored in a neutral medium (pH = 6.9). The stoichiometric ratio 1:1 of the complex was confirmed by the A L solubility diagram according to the Higuchi and Connors method, and the stability constant is K-s = 583.38 M-1 at 25 …

Inclusion complex of tramadol in β-cyclodextrin enhances fluorescence by preventing self-quenching

Fluorescence self-quenching occurs at high concentration. Inhibition of self-quenching by inclusion of fluorescence emitters
inside the hydrophobic cavity of β-cyclodextrin (β-CD) has been addressed taking the example of the fluorescence behavior
of Tramadol hydrochloride. Indeed complexation by β-CD enhanced fluorescence emission of Tramadol under conditions
where self-quenching was operative. A quantitative account of self-quenching and its inhibition by β-CD was done through
determination of complexation equilibrium by 1H NMR experiments and a detailed study of absorption and fluorescence properties.
Tramadol and β-CD associate as a complex of 1:1 stoichiometry with a formation constant K11 = 260. Complexation of
Tramadol by β-CD does not cause modification of its absorbance and fluorescence spectra. Fluorescence self-quenching of
Tramadol above ∼ 1 mmol·L−1 was characterized by a Stern–Volmer constant K = 810 L·mol−1. Inhibition of self-quenching
by formation of an inclusion complex was manifested by lower Stern–Volmer constants in the presence of β-CD. Such study
required a correct account of Inner Filter Effects on fluorescence, which is mandatory in all physicochemical studies using
fluorescence where concentrations are rather high.

Investigation of cyclodextrin inclusion compounds using FT-IR, SEM and X-ray diffraction

The prepared complexes of inclusion are characterized by IR where we have deduced the presence of new liaisons, this we permitted to say that, it has a formation of a new bands and we also concluded that the intensity of absorption of the majority of the frequencies are moved in the complexes which are weaker than in pure Tramadol. In the literature an amorphous material is more reactive than a crystalline one and as consequence amorphous materials are considered more hygroscopic when compared to crystalline solids. The results obtained by X-rays shows a crystalline semi profile for the complexes prepared by the three methods contrary to Tramadol and β-CD which present a crystalline profile. Modification of crystals and powder can be also observed by SEM and assumed as a proof of the formation of new inclusion complex.

A novel method for the deprotection of N-Boc-D-glucosamine-Ac by irradiation via microwave

1986 in the field of the organic synthesis, the reactions assisted by microwaves are used more and more in the strategies of synthesis. It meets an interest growing because of its great effectiveness and its extreme profitability due to reduced reaction times, better outputs and obtaining purer products. Its particular use, was the subject of a method around whose developed a green chemistry. Several developed, he was devoted, and the last in date is a work CO Kappe in 2005. Within the framework of this work, we will try to develop an aspect of the chemistry of the protective groupings: chimioselective protection deprotection.

First total syntheses of (±)-3-aza-11-selena and (±)-3-aza-11-tellura steroids

An efficient synthesis of 11-selena and 11-tellura steroids bearing a pyridine as an A ring was achieved via an intramolecular Diels–Alder cycloaddition of o-quinodimethanes, which were generated from a 3-azabicyclo[4.2.0]octa-1,3,5-trien-7-one ketal. The major isomer matches the trans-anti-trans ring configuration of natural products. Finally, the vinyl groups of the synthesized 11-hetero steroids have been oxidized by the Wacker process in good yields. The characteristic 1H and 13C NMR spectroscopic features of the synthesized compounds are reported.

New Pseudonucleosides Containing Chiral Oxazolidin-2-Ones and Cyclosulfamides as Aglycones: Synthesis and Antiviral Evaluation

A series of chiral cyclosulfamides and oxazolidinon-2-ones have been synthesized starting from aminoacids. Regioselective substitution of these pseudopyrimidic heterocyles was carried out under Mitsunobu conditions. Best substitution results were obtained by preliminary deprotection of cyclosulfamides and their condensation with β -D-ribofuranose. Chiral oxazolidin-2-ones were coupled directly with D-ribofuranose. All compounds were tested against HSV-2, VV and SV viruses. Two compounds 6b and 6e showed significant activities against HSV-type 1.

A novel approach toward the synthesis of 1,3,4,6-tetra-O-Acetylazomethine based on D-glucosamine

A novel method was developed for synthesizing a series of 1,3,4,6-tetra-O-acetylazomethin-B-D- glucosamine (5) starting from hydrochlorid β-D-glucosamine (1) with three different aldehydes in the presence of 15% sodium hydroxide as catalyst. In these reactions 1/1, the condensation product as
half-unit ligand was obtained. The attractive features of this protocol are use of an inexpensive catalyst, operational simplicity, short reaction times, easy handling, and good yields. Mild conditions, simplicity and easier work-up are the main advantages of this method. In parallel, in this work we succeed to separate the two diastereoisomers of products obtained (5a) which are characterized by Visible UV and separated by crystallization to give (E)-isomer and (Z)-isomer.

Développement d’une méthode d’extraction de la Phycocyanine à partir de la Spiruline "Arthrospira Platensis", et l’étude de la Stabilisation par formation d’un Complexe avec la β-Cyclodextrine

La Phycocyanine (PC) est le principal pigment de la cyanobactérie "Spiruline :ArthrospiraPlatensis". La PC est une phycobiliprotéinenon-toxique, hydrosoluble, bioactive (anti-inflammatoires, anti-tumorales, anti-oxydantes…etc).La PC semble 6000 à 7000 fois plus actif que la spiruline elle-même et 20 fois plus puissante que la vitamine C. C’est une molécule de grand intérêt notamment en raison de ses propriétés bénéfiques pour la santé humaine. Par conséquent, la PC est un pigment bleu très instable car elle est sensible à la lumière, à la température et au milieu basique où elle se dégrade rapidement, ce qui rend leur utilisation limitée ; pour cette raison, nous avons étudié sa stabilité par l’ajout d'un stabilisant. Nous avons choisi la β-Cyclodexterine (β-CD).Cette dernière est molécule naturellenon toxique, hydrosoluble, biocompatible et biodégradable, permettant ainsi leurs utilisations dans des domaines allant de la pharmaceutique à la médecine et à
l'agriculture. La β-CD est déjà largement utilisée dans le milieu pharmaceutique, principalement comme adjuvant de solubilisation pour augmenter la stabilité et/ou améliorer la solubilité et elle peut protéger la photodégradation de certaines substances actives. Ce travail est d’abord conduit au développement d’une méthode d‘extraction assistée par ultrasons consiste en une macération dans le mélange (glycérol/eau) avec différentsrapports molaires de glycérol et d’eau (1/9, 2/8, 3/7, 4/6 et 5/5), suivi par une précipitation au sulfate d'ammonium. Il a pu être démontré que cette méthodologie permet d‘extraire rapidement (15min)et efficacement de la PC à partirde la Spiruline ''Arthrospira Platensis'' (CCC540) déjà cultivé dans notre laboratoire avec un rendementde 52.93mg/g, la concentration en PC est 3.93mg/mL etle degré de pureté est 0.79 (dans le cas de 2/8).Enfin, l’étude de la stabilité de la PC par la formation d’un complexe avec la β-CD montre que la PCest devenu plus stable après l’ajout d'un stabilisant et nous avons pu la conservé pendant une longue durée.

Action Antioxydant, Antinitrosante et antibactérienne de l’Acide Syringique dans des milieux modéles.

L’objectif de cette étude était dévaluer des méthodes d’extraction de l’acide syringique (AS) dans le but de pouvoir déterminer la méthode optimale en termes de rendement et de l’encapsuler (AS) dans l’oligosaccharide cyclique (β-cyclodextrine) (β-CD) en vue de le stabiliser, pour ayant plus d’applications potentielles. Nos travaux ont été portés tout d’abord, sur l’optimisation pour extraire l’AS, à partir de la datte Deglet-Nour (Phoenix dactylifera) à l'aide d'ultrasons couplés à une hydrolyse thermique à 100°C. En conséquence, nous avons réussi à remettre le développement d’une stratégie modèle d'extraction de composé cible plus respectueuse de l’environnement et avec un excellent rendement (82%). Nous suggérons que cette augmentation du rendement est principalement due à l’hydrolyse thermique. En parallèle, nous avons essayé de stabiliser l’AS par la formation d’un complexe d’inclusion (1:1) avec la β-CD pour le conserver et le rend plus stable. L’étude de l’activité antioxydant et l’activité antibactérienne de l'AS libre et en complexe ont montré que, l’encapsulation a conservé les propriétés biologiques et que le complexe d’inclusion (β-CD:AS) (1:1) peut être considéré comme outils prometteur pour l’optimisation de la formulation alimentaire et
pharmaceutique.

Characterization of Inclusions complexs D-Glucosamine derivatives with β-CD

In this Work, we were interested on the synthesis of D-glucosamine derivatives. Within two steps; Acylation of the 1,3,4,6-tetracetyl-amino-2-deoxy-β-D-glucosamine hydrochloride (2) and the regioselective substitution by thiophenol. Given that this compound has very poor aqueous solubility and to ameliorate this, we have synthesized an inclusion complex between derivative (5) and β-CD, the preparation of the complex was carried out by two different methods (Physical mixture method and Solvent evaporation method).The results of this work were confirmed by structural characterization by spectroscopic methods 19F NMR, 1H NMR and IR.Finally we studied the solubility of derivative (5) in an aqueous solution containing various concentrations of β-CD by UVVisible spectroscopy which was deducted enhancement of the derivative (5) solubility in the presence of β-CD.

Obtaining Of Native Glucosamine From Food Waste: Extraction, Characterization & Biological Evaluation

Chitin and Chitosan are polysaccharide polymers containing more than 5000 gucosamine and Acetyl-glucosamine units, respectively, and their molecular weights are over one million Daltons. Glucosamine is an amino sugar and an important precursor in the synthesis of glycosylated proteins and lipids. Glucosamine is a natural bioactive substance obtained from waste co-products of shrimp waste, corn powder and peanut shells and through a chemical process involving demineralization, deproteinization and deacetylation. Although it is a common dietary supplement, there is little evidence of its effectiveness in relieving arthritis or pain. The purpose of this study was to optimize and evaluate the production conditions of glucosamine and to characterize them. Experiments have focused on the effects of interactions of five factors. Our result is made on the study of bibliographies in the same context. From the synthesis of the bibliography results, it can be concluded that: The optimal values obtained experimentally demineralization with a concentration of HCl (1.5N), agitation temperature (50 to 80°) for (2 to 3h). The highest degree deacetylation (DD) value (88%) under optimized condition of (70%) sodium hydroxide at temperature (121-122°C) for (3-4 h). The evaluation of the antioxidant activity of chitin, chitosan and native glucosamine was studied using several in vitro tests: trapping activity against the radical DPPH. Results indicate that chitosan reflects higher antioxidant potency compared to chitin and glucosamine.
